Global Glycosylation Analysis

We collaborate with a prestigious research institute to perform glycosylation analysis for various samples. We analyze purified glycoproteins, cells, cell membrane extracts, cell lysates, and tissues. The most requested glycosylation analysis is global N- and O-linked glycan profiling. Structural analysis of N- and O-linked glycans involves multiple steps.

  1. N-glycans and/or O-glycans are released from the samples by enzymatic or chemical method (e.g., PNGase F, β-elimination).
  2. In some cases, the released glycans are derivatized (e.g., methylation) prior to MS analysis.
  3. The glycans are then subjected to MALDI-TOF or ESI-MS analysis.

In addition, we provide N- and/or O-glycan profiling by labeling reagents, such as 2-aminobenzamide (2-AB), 2-aminobenzoic acid (2-AA). The labeled glycans can be subjected to subsequent HPLC or LC/MS analysis.

Glycoproteomics Analysis (site-specific analysis)

Sometimes, scientists want to characterize a target protein’s glycosylation, including N- and O-glycoform structures. In this case, we can map glycosylation sites and run a glycan analysis at each site. Site-specific glycosylation analysis involves multiple steps.

  1. The peptides or glycopeptides are prepared by protease digestion of samples.
  2. The digested samples are cleaned and subjected to LC-ESI-MS/MS or nano-LC-NSI-MS/MS. The Electron transfer dissociation (ETD) and high collision dissociation (HCD) fragmentation capabilities enhance glycopeptide analysis.
  3. The MS analysis software is used for the interpretation of the MS data.

Glycosyl Linkage Analysis by GC-MS

For this type of analysis, the polysaccharides or oligosaccharides are derived by methylation, hydrolysis, reduction, and acetylation before GC-MS analysis.
